It's common for users to make mistakes during this critical step, which can significantly affect the accuracy of their results. The washing step is essential because it removes unbound antibodies that may cause background noise, thereby improving the signal-to-noise ratio of the assay. Proper washing between each incubation ensures only specific antibody-antigen interactions are retained, leading to clearer and more reliable results. If washing is not done effectively, you might end up with high background levels or inconsistent readings, which can compromise your entire experiment.
When using an automated washer, two key factors influence the effectiveness of the washing: the volume of wash solution used and the number of wash cycles. While more washes typically lead to lower background, too many can reduce the signal strength, making it harder to detect the desired result. A common practice is to perform three washes after each incubation step. However, it’s always a good idea to follow the manufacturer’s guidelines, as some ELISA plates may require fewer washes than others.
For user-prepared plates, optimizing the washing protocol is especially important. One effective way to enhance washing efficiency is by using an excess amount of wash solution. For a 96-well plate, each well can hold around 330–460 μl of liquid. Some automated washers allow you to dispense more than that, such as 1 ml per well. The trick here is to activate both the dispensing and aspiration functions simultaneously. This means while the washer adds more liquid, it also draws it back out, ensuring thorough cleaning without spilling into adjacent wells.
